Histological Method for Determining Freshness

 The method for determining the freshness of meat using histological sections is based on the study of its microstructural characteristics, namely: the condition of the nuclei of muscle fibers, the condition of the longitudinal and transverse striations of muscle fibers, the localization of microflora, and the boundary of its distribution.

To determine the degree of freshness of meat, the microstructural characteristics of histological sections of experimental samples are assessed.In addition, using a microscope, the presence of cell nuclei of animal tissue, which should be dark blue, and cytoplasm, which should be red in varying intensity and shade, is determined in the sections. The microstructural state of the nuclei of muscle fibers, their transverse and longitudinal striations, the localization of microflora, and the limits of its spread are also evaluated.

Required equipment:

1. Biological microscope

2. Eyepiece micrometer

Represents a microscope eyepiece with a scale applied to the glass surface. A micrometer eyepiece is used to determine the relative sizes of objects in the microscope's field of view, and in combination with a stage micrometer, to determine the absolute sizes of objects under the microscope. Thanks to standard eyepiece sizes, they can be used with microscopes from different manufacturers. Important characteristics when selecting a micrometer eyepiece include the mounting diameter, magnification, scale length, and scale division. The mounting diameter of an EUROMEX micrometer eyepiece can be 30 mm or 23.2 mm.


3. Stage micrometer

This is a microscope slide with an applied scale, having an absolute dimensional value. Micrometer objects are used to determine the scale division value of an eyepiece micrometer depending on the objectives used (magnification levels). They are also used to calibrate a microscopic camera when measuring the sizes of objects using software.
